    Holocene Sea Level Recorded by Beach Rocks at Ionian Coasts of Apulia (Italy)

    Beach rocks are located along many coasts of the Mediterranean basin. The early diagenesis environment and the mean sea level along the shoreline make these landforms useful in the reconstruction of relative sea-level changes and, in particular, as SLIPs (sea-level index points). The beach rocks surveyed along the Ionian coast of Apulia were found to be well preserved at three specific depth ranges: 6–9 m, 3–4 m, and from the foreshore to about 1.20 m. Morpho-bathymetric and dive surveys were performed to assess both the geometries and the extension of the submerged beach rocks. Samples were collected at these different depths in the localities of Lido Torretta, Campomarino di Maruggio, San Pietro in Bevagna, and Porto Cesareo. Bivalve shells were identified and isolated from the beach rock samples collected at a depth of 7 m; AMS dating provided a calibrated age of about 7.8 ka BP. Their morphology and petrological features, along with the time constraints, enabled us to (i) reconstruct the local sea-level curve during the Holocene, (ii) corroborate acquired knowledge of the relative sea-level history, and (iii) identify possible local vertical land movement (VLM)

    Impact of debris flows on filter barriers: Analysis based on site monitoring data

    Debris flows are one of the most complex and devastating natural phenomena, and they affect mountainous areas throughout the world. Structural measures are currently adopted to mitigate the related hazard in urbanized areas. However, their design requires an estimate of the impact force, which is an open issue. The numerous formulae proposed in the literature require the assignment of empirical coefficients and an evaluation of the kinematic characteristics of the incoming flow. Both are generally not known a priori. In this article, we present the Grand Valey torrent site (Italian Alps). A monitoring system made up of strain gauges was installed on a filter barrier at the site, allowing the evaluation of impact forces. The system provides pivotal information for calibrating impact formulae. Two debris flows occurred during the monitoring period. We present the interpretation of videos, impact measurements, and the results of numerical analyses. The combined analysis allows a back calculation of the events in terms of forces, flow depth, and velocity. Thus, we investigate the applicability of the impact formulae suggested in the literature and of the recommended empirical coefficients. The results highlight that hydrostatic effects dominated the impact during the first event, while hydrodynamic effects prevailed in the second one

    Effect of stoned olive pomace on rumen microbial communities and polyunsaturated fatty acids biohydrogenation: an in vitro study.

    Background The stoned olive pomace (SOP), which represents approximately 50% of the conversion process of olive in oil, is largely not-utilized and creates costs for its disposal and problems related to environmental impact. In-vitro trial experiments were employed to study the effect of feeds integrated with this bio-waste rich in polyphenols on rumen biohydrogenation, using sheep rumen liquor as inoculum. Results Fatty acid (FA) analysis and polymerase chain reaction denaturing gradient gel electrophoresis (DGGE) approach aimed to microbial community characterization showed that including SOP in feeds at the level of 50 g/kg and 90 g/kg induced changes in FA profile and in microbial populations. The contemporary decrease of Butyrivibrio proteoclasticus and the accumulation of vaccenic acid was observed. A depression of Neisseria weaveri, Ruminobacter amylophilus and other unclassified bacteria related to members of the Lachnospiraceae and Pasteurellaceae families was detected, suggesting that these microbial groups may be involved in rumen biohydrogenation. Conclusions Supplementation of feeds with SOP changes rumen bacterial community, including bacteria responsible for vaccenic acid hydrogenation to stearic acid, thus modifying FA profile of rumen liquor. Hence, an use of SOP aimed to produce meat or dairy products enriched in functional lipids could be hypothesized

    Intrinsically active MEK variants are differentially regulated by proteinases and phosphatases

    MAPK/ERK kinase (MEK) 1/2 are central signaling proteins that serve as specificity determinants of the MAPK/ERK cascade. More than twenty activating mutations have been reported for MEK1/2, and many of them are known to cause diseases such as cancers, arteriovenous malformation and RASopathies. Changes in their intrinsic activity do not seem to correlate with the severity of the diseases. Here we studied four MEK1/2 mutations using biochemical and molecular dynamic methods. Although the studied mutants elevated the activating phosphorylation of MEK they had no effect on the stimulated ERK1/2 phosphorylation. Studying the regulatory mechanism that may explain this lack of effect, we found that one type of mutation affects MEK stability and two types of mutations demonstrate a reduced sensitivity to PP2A. Together, our results indicate that some MEK mutations exert their function not only by their elevated intrinsic activity, but also by modulation of regulatory elements such as protein stability or dephosphorylation
